Hybridoma Development

Hybridoma development is a multiphase process that begins with an in depth consultation where we spend time understanding your needs so an appropriate development strategy can be applied to increase your chances of obtaining an antibody that satisfies your goals. Multiple factors are considered  as we create your custom hybridoma development program:

  • Your final assay format such as Elisa, Flow Cytometry, Western blot, IHC, and Immunofluorescence
  • Custom antigen design and production if required
  • Custom immunization strategies using a variety of mouse strains
  • Screening methods compatible with your end use.

Once a strategy is developed the process moves into the first phase where our expert management team at SDIX will provide you with exact time lines for every step of the process so you can manage your goals most effectively.

 

Phase I: Antigen production and Immunization

  • Antigen production
  • A minimum group of 5 mice will be immunized over a period of 11 weeks. Strains of mice available are Swiss Webster and Balb/c. Please contact us for additional strains .
  • Serum is screened by EIA or an alternative custom strategy
  • Immune serum is delivered to the client for evaluation with a customized data package
  • Technical discussion with client and SDIX technical team to review data
  • Selection of mice to move into phase II
  • Timing ~12 weeks

Animals chosen for their unique serum antibody activities will be employed for the development of hybridomas (phase II).

 

Phase II: Cell Fusion, Screening and Cloning

  • Fusion of mice to myeloma cell line
  • Hybrids are screened by chosen method at 96 well and 24 well level. Deliver positive supernatants to client for evaluation
  • Client selects parental hybrids to be frozen down (5 vials each)
  • Client selects hybrids for subcloning and freezing (10 vials each)
  • Deliver 75ml supernatant from each subclone
  • Delivery of final project summary report
  • Timing ~12 weeks

After a stable hybridoma is created and subcloned the monoclonal production phase III can begin.

 

Phase III: R&D Small Scale Production

  • Perform a 10 mouse ascites or 3.0L roller bottle run with Protein A Purification, on a SDIX generated subclone developed during a Hybridoma Development Project
  • Timing ~8 weeks
